Enzymes, Active Site & Substrate

Enzymes: Unlocking the Mystery of Active Sites and Substrates

Enzymes are like tiny machines in your body, speeding up essential chemical reactions. But how do they know what to work on? That’s where the active site and substrate come in

Active Site and Substrate: A Perfect Match

Key Terms:

Active Site:

  • A special pocket or groove on the enzyme where the reaction happens.
  • It has a unique shape that matches a specific molecule.

Substrate:

  • The molecule an enzyme works on.
  • It fits into the active site like a puzzle piece.

The Lock-and-Key Model

Think of the enzyme and substrate as a lock and key:

  • Lock (Enzyme): The active site is the lock, perfectly shaped for one specific key.
  • Key (Substrate): The substrate fits perfectly into the active site.

If the key doesn’t fit, the lock won’t open—this is why enzymes are so specific

How Do Enzymes Work?

Step-by-Step Process:

Binding:

  • The substrate binds to the enzyme’s active site, forming an enzyme-substrate complex.

Reaction:

  • The enzyme helps break down or build up the substrate into products.

Release:

  • The products are released, and the enzyme is free to work again.

Example:

  • Enzyme: Amylase (found in saliva).
  • Substrate: Starch.
  • Product: Simple sugars like maltose

What Makes Enzymes Specific?

  1. The active site has a unique shape that only fits a specific substrate.
  2. This is called enzyme specificity—only the right key (substrate) can activate the lock (enzyme).

Lock-and-Key Rule: The substrate must fit perfectly into the enzyme’s active site.

Optimum Conditions Matter: Enzymes work best at specific temperatures and pH levels.

Reusable Machines: Enzymes are not used up—they can keep working after the reaction
